I have a 33 gallon aquarium that I want to drill the bottom of. It looks to me like 3/8". I want to put something big like 1.5 - 2 in bulkheads. If the bottom pane is supported, can I get away with it?

Be REAL sure... Most bottoms are tempered (though in a 33 your chances are better than it would be with a larger tank). Most sides and backs are not. The back would be a safer route. but the safest is to see if you can find out over the net (i.e. the manufacturers site)
