I am setting up a 40g Breeder as a sump and in order to get it under the stand, I had to remove the plastic frame.

1- Is the lack of the frame something to be concerned with?

2- I assume I should not rest it directly on the wood base of the stand, what should I put between the 40g and the wood stand?

You could cut a piece of closed cell foam. Like a yoga mat. To go under to help support the weight / distribute & cushion.. Keep in mind also that traditional glass tanks with black rim (top & bottom) are usually there for structural support.

1- Yes its a concern. These tanks are made with thin glasses and only so much glue to hold the seams together. The plastic trim is to help hold it all together. Will it fail without it? Maybe, maybe not. But there is a good chance it could, so that's entirely up to you if you want to take that risk.

2- I'm assuming you removed the trim from the top and bottom. So, as aquaworld75 mentioned, yoga mat works great for this purpose.

I would recommend a leak detector of some sort.
