Who to trust? Alkalinity

So, my alkalinity test requires 5ml of water. No biggie... Except, for some reason my alkalinity was measuring low. I battled and battled... then one day I decided to use my 5 ml syringe to fill the test tube and what did I find? The 5 ml line on the test tube was significantly lower than the 5 ml of water I just measured out using the syringe. So my question is... do I trust/run the test going off the test tube marker or do I measure out 5 ml of water with the syringe? Test results are significantly different depending on the amount of test water. Thoughts?
 
I would trust your measurement. A line on a test tube is nothing more then a stamp and could have gotten off in a machine. where as a syringe is made for handling more important tasks like medication for humans.
